Comparison of Molecular Weight Distribution of Commercially Purified Oat and Barley ¥â - glucans

Commercial oat and barley ¥â-glucans were fractionated by gel permeation chromatography (GPC). Total ¥â-glucan contents (1-3), (1-4)-¥â-D-glucopyranosyl of each fraction, measured by flow injection analysis (FIA) and by the anthrone-sulfuric acid (ASA) method for total carbohydrates, were compared, and molecular weight (MW) distributions of oat and barley ¥â-glucans were observed. Barley ¥â-glucan showed two defined MW distributions of 9.8¡¿10^7 to 3.4¡¿10^7 g/mole and 2.7¡¿10^5 to 2.7¡¿10©øg/mole. The observed MW range for oat ¥â-glucan was about 4.0¡¿10^6 to 2.0¡¿10©ùg/mole. At a ¥â-glucan concentration of below 0.02 §·/§¢, the FIA was not as sensitive as the ASA method.
